SDS binds to proteins (approx 1.4g SDS /1.0g proteins), denaturates them and confers a negative charge to proteins.

A tracking dye (such as bromophenol blue) is usually added n order to follow sample migration through the gel.
If reducing conditions are required, DTT or 2-mercaptoethano may be added to the sample prior to separation.
