DNA ligation using the T4 DNA Ligase

Posted by: dami82, UIC, Chicago, IL, http://www.biotechworld.it/portfolio/fantini

The ligation of DNA fragments is a key step in many molecular biology applications, in particular in molecular cloning procedures. DNA ligases catalyze the formation of a phosphodiester bond between the 3^ hydroxyl and 5^ phosphate of adjacent DNA residues. Always KEEP the T4 ligase at -20°C (or on ice).
