The restriction digest of plasmid DNA is a molecular biology procedure aimed at cutting a plasmid DNA at specific sites using restriction endonucleases. The restriction digest is usually performed with an excess of enzyme (10U to 20 U per 1 μg plasmid) in a total volume of 50 μL at 37C for 1h. If required, the reaction may be scaled up or down, but it is important NOT TO change the ratio DNA/enzyme/total volume.
